The GoldiBlot™ process: Ni-NTA-Nanogold® binds to a polyhistidine (His) tag, then gold enhancement generates visible signal.


Features and Advantages

  • Economical: No expensive primary or secondary antibodies, or chemiluminescent or chromogenic substrates. Cost per blot is only about $22 (USD; based on 10 mL reagent volume per blot, or about one 7 x 8.4 cm blot).
  • Fast: Visualization is complete in about an hour. In contrast, protocols that use antibodies take about three hours to complete.
  • Universal: GoldiBlot™ detects His-tags regardless of position: C-terminal, N-terminal, or even internal. GoldiBlot™ can detect His-tagged fusion proteins that are sometimes undetectable with anti-His-tag antibodies.
  • Sensitive: the Ni2+- NTA interaction is as strong as antibody-antigen binding (binding constant ~1013 M-1), providing strong, specific binding at low concentrations and yielding clear, sensitive visualization.
  • Reliable: while the Ni2+- NTA interaction is as strong as antibody-antigen binding, it is a simple reaction that avoids the variability of anti-His tag antibodies.
  • Highly Specific: Ni2+ - NTA Nanogold® nanoparticles have similar binding capabilities to IMAC, and specifically detect polyhistidine tags without any requirement for specific adjacent groups.
  • Direct Visualization: No chemiluminescent substrates, X-ray film or CCD imagers.
  • Stable: all reagents are highly stable, with shelf life longer than three years.
  • Long-lived high contrast signal: dark purple-black staining is stable, long-lived and does not fade, making imaging and archiving straightforward.
